运用多聚酶链扩增反应(PCR)及瑞粒重复扩增方法(TRAP)法检测肿瘤细胞Hela、A431和KB在电离辐射后hEST2功能区片段表达和瑞粒酶活性的变化。结果表明:肿瘤细胞hEST2功能区片段表达和端粒酶活性呈剂量依赖性下降;hEST2功能区片段的PCR法测定结果与TRAP法相符合;提示电离辐射对瑞粒酶活性的抑制是肿瘤放射治疗的重要机制之一。端粒酶活性检则可能成为评估电离辐射对肿瘤杀伤作用的新指标。
The expression of hEST2 functional fragment and the activity of telomerase activity of tumor cells Hela, A431 and KB after ionizing radiation were detected by polymerase chain reaction amplification (PCR) and repeat repeat amplification assay (TRAP). The results showed that the expression of telomerase activity and hTERT2 protein expression in tumor cells decreased in a dose-dependent manner. The results of PCR assay of hEST2 functional fragment coincided with the TRAP method. It was suggested that the inhibition of telomerase activity by ionizing radiation was inhibited by tumor radiotherapy One of the important mechanisms. Detection of telomerase activity may be a new indicator of the effect of ionizing radiation on tumor killing.
