目的研究免疫抑制剂来氟米特对兔动脉粥样硬化形成的影响。方法24只新西兰大白兔随机分为三组:正常饮食组、高脂饮食组和高脂饮食+来氟米特治疗组。在0周和8周时测定血清中血脂、肿瘤坏死因子α、白介素介素6浓度。免疫组织化学染色测定斑块处巨噬细胞数量。图像分析观察主动脉斑块内膜面积比。结果来氟米特对血脂无明显影响,但可使肿瘤坏死因子α、白介素介素6、主动脉斑块内膜面积比及斑块处的巨噬细胞数较高脂饮食组显著降低(P均<0.001)。结论来氟米特可显著抑制动脉粥样硬化部位的炎症反应,对兔动脉粥样硬化有预防作用。
Objective To study the effect of leflunomide, an immunosuppressive drug, on the development of atherosclerosis in rabbits. Methods Twenty-four New Zealand white rabbits were randomly divided into three groups: normal diet group, high fat diet group and high fat diet + leflunomide treatment group. Serum lipids, tumor necrosis factor alpha and interleukin 6 concentrations were measured at 0 and 8 weeks. Immunohistochemical staining was used to determine the number of macrophages at the plaque. Image analysis of aortic plaque intimal area ratio. Results Leflunomide had no significant effect on blood lipids but significantly decreased tumor necrosis factor-α, interleukin-6, aortic plaque intima-to-plaque area and macrophage count at plaque All <0.001). Conclusion Leflunomide can significantly inhibit the inflammation of atherosclerotic parts of the rabbit atherosclerosis have a preventive effect.
