目的:考察不同型号大孔吸附树脂对莱菔子水提液的纯化效果。方法:采用HPLC测定芥子碱硫氰酸盐含量,流动相乙腈(A)-0.1%磷酸(B)进行梯度洗脱(0~15 min,5%~10%A;15~17 min,10%~12.5%A;17~27 min,12.5%~14%A;27~55 min,14%~25%A;55~57 min,25%~70%A;57~70 min,70%~100%A),检测波长225 nm。以芥子碱硫氰酸盐洗脱率和HPLC指纹图谱共有峰的总有效成分洗脱率为指标,比较各型号树脂纯化效果,筛选最佳型号大孔树脂。结果:各树脂对芥子碱硫氰酸盐洗脱率的影响顺序为AB-8>NKA-9>HPD100C>HPD400>X-5>HPD750>HPD722>D101>DM130,对总有效成分洗脱率的影响顺序则为HPD400>HPD100C>HPD722>AB-8>NKA-9>HPD750>DM-130>D101>X-5。AB-8型树脂对芥子碱硫氰酸盐的洗脱率达96.39%,对总有效成分的洗脱率70.03%。结论:采用AB-8型大孔吸附树脂纯化莱菔子水提取液有效成分是可行的。
Objective: To investigate the purification effect of different types of macroporous resin on the water extract of Radish. Methods: The content of sinapine thiocyanate was determined by HPLC. The mobile phase consisted of gradient elution of acetonitrile (A) -0.1% phosphoric acid (B) (0-15 min, 5-10% A, 15-17 min, ~ 12.5% A; 17-27 min, 12.5-14% A; 27-55 min, 14-25% A; 55-57 min, 25-70% A; 57-70 min, 70-100% % A), detection wavelength of 225 nm. The elution rate of total erucic acid thiocyanate and the common peak of HPLC fingerprinting were used as indexes to compare the purification results of different types of resin and the best macroporous resin was screened. Results: The order of the elution rate of thiocyanate for each resin was AB-8> NKA-9> HPD100C> HPD400> X-5> HPD750> HPD722> D101> The order of impact is HPD400> HPD100C> HPD722> AB-8> NKA-9> HPD750> DM-130> D101> X-5. The elution rate of AB-8 resin to sinapine thiocyanate was 96.39%, and the elution rate of total active ingredient was 70.03%. Conclusion: It is feasible to purify the effective components of the aqueous extract of Radish by using AB-8 macroporous adsorption resin.
