目的:通过正交实验确定芪术颗粒剂中黄芪甲苷的最佳提取工艺;建立黄芪甲苷的含量测定方法。方法:色谱柱:Prodigy 5u C18色谱柱(250 mm×4.6 mm,5μm)。流动相:乙腈-水(35∶65)。流速:1.0 mL/min。柱温:24℃。ELSD参数:飘移管温度100℃,氮气流速1.5 mL.min-1。结果:精密度试验RSD为1.59%,稳定性试验RSD为1.42%;重复性试验RSD%为2.67%;加样回收率试验结果平均回收率为98.10%,RSD为0.87%。结论:该提取工艺稳定,提取率高。该含量测定方法的分离度高,重现性好,获得了满意的结果,可作为控制产品质量的指标之一。
OBJECTIVE: To determine the best extraction technology of Astragaloside IV from Qizhu Granules by orthogonal experiment. To establish a method for the determination of Astragaloside IV. Method: Column: Prodigy 5u C18 column (250 mm × 4.6 mm, 5 μm). Mobile phase: acetonitrile-water (35:65). Flow rate: 1.0 mL / min. Column temperature: 24 ℃. ELSD parameters: drift tube temperature 100 ℃, nitrogen flow rate 1.5 mL.min-1. Results: The precision test RSD was 1.59%, the stability test RSD was 1.42%, the repeatability test RSD% was 2.67%. The average recovery rate was 98.10%. The RSD was 0.87%. Conclusion: The extraction process is stable and the extraction rate is high. The method has the advantages of high resolution, good reproducibility and satisfactory results, which can be used as an index to control product quality.
