目的 研究大肠肿瘤中细胞核形态参数和 p5 3、PC NA定量表达的关系。方法 应用免疫组化和图像分析技术对 40例腺瘤、80例腺癌及 15例正常粘膜细胞核形态参数和 p5 3、PCNA含量进行研究。 结果 腺瘤及腺癌中核面积、核周长、核平均直径均大于正常粘膜 (P <0 0 5 ,P <0 0 1) ,腺癌组 p5 3及PCNA相对含量均高于腺瘤组 (P <0 0 5 )。PCNA表达与大肠癌的差分化有关 (P <0 0 5 )。核面积及核平均直径与腺瘤及腺癌中PCNA及p5 3含量正相关 ,与腺瘤及腺癌组织学类型无关。结论 检测大肠肿瘤中核形态参数和 p5 3、PCNA相对含量有助于大肠癌早期诊断和判断预后。
Objective To study the relationship between nuclear morphological parameters and quantitative expression of p53 and PCNA in colorectal tumors. Methods Immunohistochemistry and image analysis techniques were used to study the morphological parameters of nucleus and the content of p53 and PCNA in 40 adenomas, 80 adenocarcinomas and 15 normal mucosa. Results The nuclear area, nuclear perimeter and mean nuclear diameter of adenocarcinoma and adenocarcinoma were all higher than those of normal mucosa (P <0.05, P <0.01), and the relative contents of p53 and PCNA in adenocarcinoma were higher than those in adenoma P <0 0 5). The expression of PCNA correlated with the differentiation of colorectal cancer (P <0.05). The nuclear area and average diameter of nucleus were positively correlated with the content of PCNA and p5 3 in adenoma and adenocarcinoma, but not with the histological type of adenoma and adenocarcinoma. Conclusion Detection of nuclear morphological parameters and the relative content of p53 and PCNA in colorectal tumors is helpful for the early diagnosis and prognosis of colorectal cancer.
