目的 探讨室管膜肿瘤中白细胞介素 8(IL 8)表达和分泌的意义。方法 采用免疫组织化学法检测 5 0例手术切除的室管膜肿瘤标本中IL 8表达水平 ;双抗体夹心ELISA法测定原代培养的室管膜肿瘤细胞上清液中IL 8含量。结合患者的一般状况 ,对不同组织学分型、病理分级及侵袭性的肿瘤的IL 8表达和分泌量进行统计学检验。结果 正常脑组织IL 8表达为阴性或弱阳性 ,而室管膜肿瘤IL 8表达均为阳性。间变型或恶性室管膜瘤每高倍视野中IL 8阳性细胞数目为 417± 38个 ,多于细胞和上皮型室管膜瘤 (P <0 0 1) ;IL 8阳性染色强度为 5 6± 7 5AU ,高于细胞和上皮型室管膜瘤 (P <0 0 1)。间变型或恶性室管膜瘤细胞上清液中IL 8的含量为 6 38μg/L± 87μg/L ,高于细胞和上皮型室管膜瘤 (2 98μg/L± 48μg/L ,P <0 0 1)。侵袭性室管膜肿瘤中IL 8的表达和分泌水平高于非侵袭性室管膜肿瘤 (P <0 0 1)。高恶性室管膜肿瘤细胞中IL 8的表达和分泌高于低恶性或良性室管膜瘤 (P <0 0 1)。IL 8的表达和分泌水平与患者性别、年龄等无明显相关性。结论 高恶性室管膜肿瘤中IL 8表达和分泌显著高于低恶性或良性室管膜肿瘤细胞。IL 8的表达和分泌可能与室管膜肿瘤的发生和发展密切相关 ,可作为判断室管膜肿瘤恶性程度及患者预后
Objective To investigate the significance of interleukin 8 (IL 8) expression and secretion in ependymal tumors. Methods Immunohistochemical method was used to detect the expression level of IL-8 in 50 cases of elective ependymal tumor specimens. IL 8 level in primary culture ependymal cell supernatant was detected by double antibody sandwich ELISA. According to the general condition of the patients, IL 8 expression and secretion in different histological types, pathological grading and invasive tumors were statistically tested. Results The expression of IL 8 in normal brain tissue was negative or weakly positive while the expression of IL 8 in ependymal tumors was positive. The number of IL8-positive cells in high-power field per room or anaplastic ependymoma was 417 ± 38, more than that of cells and epithelial ependymoma (P <0.01). The positive staining intensity of IL-8 was 56 ± 7 5AU, higher than that of epithelial and ependymoma (P <0.01). The content of IL-8 in the metaphase or malignant ependymoma cell supernatant was 6 38μg / L ± 87μg / L, higher than that of the cell and epithelial ependymoma (988μg / L ± 48μg / L, P <0 0 1). The expression and secretion of IL-8 in invasive ependymal tumors were higher than those in non-invasive ependymoma (P <0.01). The expression and secretion of IL-8 in high-grade ependymoma cells was higher than that in low-grade or benign ependymoma (P <0.01). The level of IL-8 expression and secretion had no significant correlation with patient’s sex, age and so on. Conclusion The expression and secretion of IL-8 in high-grade ependymoma is significantly higher than that in low-grade or benign ependymal tumor cells. The expression and secretion of IL-8 may be closely related with the occurrence and development of ependymal neoplasms, which can be used as a criterion for evaluating the malignancy of ependymal neoplasms and the prognosis of patients
