目的:优选蛇床子挥发油羟丙基-β-环糊精(HP-β-CD)包合物的制备工艺。方法:以挥发油利用率和包合物得率为考察指标,采用正交试验法对蛇床子挥发油HP-β-CD包合物的工艺进行优化。采用UV技术、显微鉴定、GC-MS技术对包合物进行质量控制研究。结果:最佳包合工艺条件为蛇床子挥发油与HP-β-CD的体积质量比1∶8,包合时间2.0 h,包合温度40℃。采用紫外光谱法验证包合前后紫外吸收图谱基本一致,显微鉴定表明油已被包合,GC-MS验证了包合前后挥发油中各成分基本未发生变化。结论:该提取、包合工艺合理可行,为临床制剂的合理使用和包合物开发提供实验依据。
OBJECTIVE: To optimize the preparation process of the inclusion complex of the essential oil of Cnidium monnieri beta-cyclodextrin (HP-β-CD). Methods: The volatile oil utilization and inclusion compound yield as the index, Orthogonal test method for the Cnidium essential oil HP-β-CD inclusion complex process optimization. Using UV technology, microscopic identification, GC-MS technology for inclusion complex quality control study. Results: The optimum inclusion conditions were as follows: volume ratio of volatile oil to HP-β-CD was 1: 8, inclusion time 2.0 h, inclusion temperature 40 ℃. UV spectrophotometry before and after the inclusion of UV absorption spectra are basically the same, microscopic identification showed that the oil has been included, GC-MS verified before and after the inclusion of volatile oil in the basic components did not change. Conclusion: The extraction and inclusion process is reasonable and feasible, providing experimental basis for the rational use of clinical preparations and the development of inclusion compound.
