背景:内源性6-羟基多巴胺能参与氧化应激,N-乙酰半胱氨酸能有效抗氧化和清除自由基。目的:探讨6-羟基多巴胺对骨髓基质细胞的毒性作用及N-乙酰半胱氨酸对其的拮抗作用。方法:在体外培养SD大鼠骨髓基质细胞,取第3代骨髓基质细胞分别加入终浓度为0,0.05,0.1g/L的6-羟基多巴胺和终浓度为0,0.075,0.3,1.2,4.8g/L的N-乙酰半胱氨酸。结果与结论:MTT检测发现0.05和0.1g/L6-羟基多巴胺可以明显降低骨髓基质细胞的细胞活性,而加入6-羟基多巴胺的同时加入0.3g/LN-乙酰半胱氨酸可以显著抑制6-羟基多巴胺的毒性作用。提示抗氧化剂N-乙酰半胱氨酸可以影响6-羟基多巴胺的作用。
BACKGROUND: Endogenous 6-hydroxydopamine is involved in oxidative stress and N-acetylcysteine is an effective antioxidant and scavenger of free radicals. Objective: To investigate the toxic effects of 6-hydroxydopamine on bone marrow stromal cells and the antagonism of N-acetyl cysteine. Methods: SD rat bone marrow stromal cells were cultured in vitro. The third generation bone marrow stromal cells were added to 6-hydroxydopamine at the final concentrations of 0, 0.05 and 0.1 g / L, respectively, and the final concentration was 0,0.075,0.3,1.2,4.8 g / L of N-acetylcysteine. RESULTS AND CONCLUSION: MTT assay showed that 0.05 and 0.1g / L 6-hydroxydopamine could significantly reduce the cell viability of bone marrow stromal cells, while the addition of 6-hydroxydopamine and 0.3g / L-acetyl cysteine significantly inhibited the 6- Toxicity of hydroxydopamine. Tip Antioxidant N-acetylcysteine can affect the role of 6-hydroxydopamine.
