背景:缺血性脑血管病白细胞流变性与微循环密切相关,传统活血化瘀中药是否对白细胞流变学异常、变形能力下降有影响?目的:通过建立大鼠局灶性脑缺血病理模型,观察其白细胞流变性的变化,并探讨活血化瘀中药复方乌龙丹的作用。设计:随机对照的实验研究。地点和材料:实验地点:解放军第一军医大学中医系实验室。选用Wistar雄性大鼠40只,体质量(250±20)g,随机分为假手术组、模型组、乌龙丹低剂量组、乌龙丹高剂量组,每组10只。干预:大鼠结扎颈总动脉(CCA)后再阻断大脑中动脉(MCA)造模,造模术后24h取血用核孔滤膜红细胞变形能力测定仪测定白细胞变形指数、白细胞滤过堵塞的粒子浓度及红、白细胞-红细胞滤过指数。主要观察指标:实验各组大鼠白细胞总数、白细胞变形指数、白细胞滤过堵塞的粒子浓度及红、白细胞-红细胞滤过指数。结果:模型组的白细胞总数、白细胞变形指数、白细胞滤过堵塞的粒子浓度及红、白细胞-红细胞滤过指数均明显高于假手术组(P<0.01),表示白细胞的变形能力下降。乌龙丹给药组上述指标与模型组有显著性差异(F=3.761,P<0.01),显示出该方对梗死后白细胞变形能力下降有一定的对抗作用。结论:急性局灶性脑缺血损伤中有白细胞流变学异常,变形能力下降。中药复方乌龙丹对此有一定对抗作用,增强白
BACKGROUND: Leukocyte rheology of ischemic cerebrovascular disease is closely related to microcirculation. Does the traditional Chinese medicine for promoting blood circulation and removing blood stasis have an effect on the abnormality of leukocyte rheology and degeneration? Objective: To establish a rat model of focal cerebral ischemia , Observed the changes of leukocyte rheology, and discussed the role of traditional Chinese medicine compound Wulong Dan. Design: Randomized controlled experimental study. Venue and Material: Experiment Location: Laboratory of Traditional Chinese Medicine, PLA First Military Medical University. Forty Wistar male rats were randomly divided into sham-operation group, model group, low-dose Wulong Dan group and high-dose Wulong Dan group with 10 rats in each group. The body weight was 250 ± 20 g. Intervention: After the rats were sacrificed, the middle cerebral artery (MCA) was occluded by ligating the common carotid artery (CCA), and the blood was collected at 24 hours after the model was established. The leukocyte deformability index and leucocyte filtration blockage Particle concentration and red, white blood cells - red blood cell filtration index. MAIN OUTCOME MEASURES: The total number of white blood cells, leukocyte deformity index, the concentration of leucocyte filtration plugged particles and red and white blood cell - erythrocyte filtration index in each group. The above indexes in Wulong Dan administration group were significantly different from those in model group (F = 3.761, P <0.01), which showed that the prescription had a certain antagonistic effect on the deformability of leucocyte after infarction.
