目的探讨Ⅰb~Ⅱb期子宫颈癌组织中淋巴管内皮细胞体积百分比和高迁移率族蛋白1(high mobility group box1,HMGB1)表达及二者的相关性。方法采用免疫组化SP法检测26例Ⅰb~Ⅱb期中分化宫颈鳞状细胞癌、10例慢性宫颈炎和5例宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)3级手术切除标本和(或)宫颈活检标本中D2-40、HMGB1的表达,计数淋巴管内皮细胞体积百分比。HMGB1表达量化应用H评分。组织病理学量化后进行统计学分析。结果淋巴管内皮细胞体积百分比对于Ⅰb2期(8.88%±4.60%)和Ⅱb(7.75%±3.35%)期两组间差异无显著性(P=0.67),均较慢性宫颈炎(2.50%±0.61%)、CIN3(2.76%±1.24%)、Ⅰb1(2.59%±0.81%)和Ⅱa(3.09%±0.82%)期显著增高,后四组组间差异无显著性。HMGB1的表达通过H评分显示其在Ⅱa(1.07±0.54)和Ⅱb(1.09±0.29)组的表达显著增高(P=0.03);慢性宫颈炎(0.47±0.42)、CIN3(0.32±0.18)、Ⅰb(0.62±0.34)组间HMGB1表达差异无显著性。各组间淋巴管内皮细胞体积百分比与HMGB1表达无统计学相关。结论Ⅰb2和Ⅱb期淋巴管生成明显较其他各组增强,其与促炎因子和促淋巴管生成因子HMGB1的表达无明显相关性。
Objective To investigate the relationship between the volume percentage of lymphatic endothelial cells and the expression of high mobility group box 1 (HMGB1) in stage Ⅰb ~ Ⅱb cervical cancer. Methods Immunohistochemical SP method was used to detect grade Ⅰb ~ Ⅱb differentiated cervical squamous cell carcinoma, 10 cases of chronic cervicitis and 5 cases of cervical intraepithelial neoplasia (CIN) Cervical biopsy specimens of D2-40, HMGB1 expression, counting the volume percentage of lymphatic endothelial cells. HGB scores were evaluated using HMGB1 expression quantification. Histopathology was quantified for statistical analysis. Results The volume percentage of lymphatic endothelial cells showed no significant difference between the two groups (χ2 = 0.67, 8.88% ± 4.60%) and Ⅱb (7.75% ± 3.35% %), CIN3 (2.76% ± 1.24%), Ⅰb1 (2.59% ± 0.81%) and Ⅱa (3.09% ± 0.82%) respectively. There was no significant difference between the latter four groups. The expression of HMGB1 was significantly higher in the groups of IIa (1.07 ± 0.54) and IIb (1.09 ± 0.29) by H score (P = 0.03); the chronic cervicitis (0.47 ± 0.42), CIN3 (0.32 ± 0.18) (0.62 ± 0.34) group HMGB1 expression difference was not significant. There was no significant correlation between the volume percentage of lymphatic endothelial cells and the expression of HMGB1 in each group. Conclusions Lymphangiogenesis in stage Ⅰb2 and stage Ⅱ b is obviously stronger than that in other groups. There is no significant correlation between the expression of pro-inflammatory cytokines and lymphangiogenic factor HMGB1.
