目的探讨FAT10过表达对人乳头状甲状腺癌细胞抗凋亡及集落形成能力的影响。方法采用脂质体法将FAT10质粒导入TPC-1细胞,建立FAT10过表达的人乳头状甲状腺癌细胞,用Western blot检测细胞中的FAT10表达水平,用染色体核型分析FAT10过表达而导致的非整倍体现象,Annexin V凋亡试剂盒检测FAT10过表达对TPC-1抗凋亡能力的影响。通过集落形成实验观察其集落形成能力的变化。结果 Western blot结果显示FAT10过表达组细胞FAT10蛋白水平明显高于对照组,FAT10过表达组出现非整倍体形象,并且抗喜树碱诱导凋亡的能力和集落形成能力显著高于对照组。结论 FAT10过表达可以促进人乳头状甲状腺癌细胞抗凋亡及集落形成能力。
Objective To investigate the effect of FAT10 overexpression on anti-apoptosis and colony-forming ability of human papillary thyroid carcinoma cells. Methods FAT10 plasmid was transfected into TPC-1 cells by lipofectamine 2000, and FAT10 overexpressed human papillary thyroid carcinoma cells were established. The expression level of FAT10 in cells was detected by Western blot, and FAT10 overexpression was analyzed by chromosomal karyotype Euploid phenomenon, Annexin V apoptosis kit detection FAT10 overexpression of TPC-1 anti-apoptotic capacity. The colony formation ability was observed by colony formation experiment. Results The result of Western blot showed that FAT10 protein in FAT10 overexpression group was significantly higher than that in control group. FAT10 overexpression group showed aneuploid image, and anti-camptothecin induced apoptosis and colony-forming ability were significantly higher than those in control group. Conclusion FAT10 overexpression can promote anti-apoptosis and colony-forming ability of human papillary thyroid carcinoma cells.
