目的:探讨银杏叶提取物(GBE)抑制NF-kappa Bp65的表达在四氧化碳(CCl4)诱导Wistar大鼠肝纤维化中肝细胞损伤的意义。方法:采用CCL4诱导的大鼠肝纤维化模型。生化检测各组血清丙氨酸氨基转移酶(ALT)、天氨酸氨基转移酶(AST)。采用免疫组化S-P方法检测肝细胞NF-kappa Bp65和ICAM-1表达并分析与肝细胞损害的关系。结果:治疗组大鼠的血清肝功能指标和肝纤维化病理分级与模型组比较都有显著的改善(P<0.05);治疗组大鼠肝组织细胞因子水平与模型组比较也显著降低(P<0.05)结论:银杏叶提取物可减轻肝组织的损伤及其纤维化程度,通过抑制NF-kappa Bp65的表达,进一步减弱了ICAM-1的表达,可能是其抗肝纤维化作用的靶点之一。
Objective: To investigate the significance of Ginkgo biloba extract (GBE) inhibiting the expression of NF-kappa Bp65 in liver cell injury induced by carbon tetrachloride (CCl4) in Wistar rat liver fibrosis. Methods: CCL4 induced rat liver fibrosis model. Biochemical detection of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST). Immunohistochemical S-P method was used to detect the expression of NF-kappa Bp65 and ICAM-1 in hepatocytes and their relationship with hepatocellular damage. Results: The serum liver function index and pathological grading of liver fibrosis in treatment group were significantly improved compared with model group (P <0.05). The level of cytokines in liver tissue of treatment group was significantly lower than that of model group (P <0.05) .Conclusion: Ginkgo biloba extract can reduce the damage of liver tissue and the degree of fibrosis, and further attenuate the expression of ICAM-1 by inhibiting the expression of NF-kappa Bp65, which may be the target of its anti-hepatic fibrosis effect one.
