利多卡因和奎尼丁在临床使用上安全范围小,易出现中毒反应。测定利多卡因及奎尼丁血浓度可有效地控制其抗心律失常作用及避免毒副作用。其测定方法主要有气相色谱法、气相色谱—质谱法、高效液相色谱法、荧光色谱和均相酶联免疫法等。国内尚未见同时测定血清中上述两药的报道,国外虽有报道,但提取方法比较复杂。作者采用提取小柱进行样品的预处理,用高效液相色谱法同时测定,为临床上监测血药浓度及研究药代动力学提供一种快速敏感和准确的方法。
Lidocaine and quinidine in the clinical use of a small security range, prone to toxic reactions. Determination of lidocaine and quinidine blood concentration can effectively control its anti-arrhythmic effects and avoid side effects. The determination of the main methods are gas chromatography, gas chromatography - mass spectrometry, high performance liquid chromatography, fluorescence chromatography and homogeneous enzyme-linked immunosorbent assay. There are no reports about simultaneous determination of the above two drugs in the serum in our country. Although it is reported abroad, the extraction method is rather complicated. The authors used extraction cartridges for sample pretreatment and simultaneous determination by high performance liquid chromatography to provide a rapid, sensitive and accurate method for clinical monitoring of plasma concentrations and study of pharmacokinetics.
