目的 :研究不同膳食组成对小鼠肌肉胰岛素受体及其相关底物基因表达的影响。 方法 :C5 7BL / 6 J小鼠分别饲以高脂饮食、高脂加谷氨酰胺饮食、高脂 3个月后补加谷氨酰胺饮食以及正常对照饮食 ,实验期为 5 .5个月。以反转录 -聚合酶链反应 (RT- PCR)分别测定肌肉胰岛素受体 (IR)及其受体底物 1(IRS- 1)、受体底物 2 (IRS- 2 )以及磷酸肌醇 3激酶 (PI- 3- kinase)m RNA水平。 结果 :高脂可使小鼠肌肉 IR,IRS- 1,IRS- 2 ,PI- 3- kinase m RNA水平不同程度下降 ,分别为正常对照的 81% ,6 3% ,5 9%及 45 %。结论 :谷氨酰胺可在一定程度上抑制上述基因 m RNA下降的趋势 ,提示营养因素对机体胰岛素敏感性的影响可能部分是通过影响其胰岛素受体及其一系列相关底物的基因表达而实现的。
Objective: To study the effect of different dietary components on the gene expression of muscle insulin receptor and its related substrates in mice. Methods: C5 7BL / 6 J mice were fed a high-fat diet, high-fat plus glutamine diet, high-fat diet for 3 months after the addition of glutamine diet and normal control diet, the experimental period was 5.5 months. The expressions of muscle insulin receptor (IR) and its receptor substrate 1 (IRS-1), receptor substrate 2 (IRS-2) and phosphoinositide 3 kinase (PI-3-kinase) m RNA levels. Results: The levels of IR, IRS-1, IRS-2 and PI-3-kinase m RNA in mice were reduced by high fat diet to 81%, 63%, 59% and 45% of normal controls, respectively. CONCLUSION: Glutamine can inhibit the decline of m RNA in the above-mentioned gene to a certain extent, suggesting that the influence of nutrition on the insulin sensitivity of the body may be partially achieved by affecting the gene expression of its insulin receptor and a series of related substrates of.
