目的:探讨环氧合酶-2抑制剂对口腔鳞癌发生过程中血管生成的影响。方法:选取人舌鳞癌细胞株Tca8113作为实验样本。应用反转录聚合酶链反应(PT-PCR)及Western印迹方法检测在COX-2抑制剂NS-398对Tca8113细胞系表达VEGF、Survivin的mRNA及蛋白水平的影响。采用SPSS11.5软件包对数据进行统计学分析。结果:NS-398可下调Tca8113细胞系VEGF及Survivin的表达,且表现出一定的时间依赖性。结论:COX-2抑制剂NS-398可降低口腔鳞状癌细胞Tca8113 VEGF及Survivin的mRNA及蛋白质含量的水平,在口腔鳞癌血管形成及基因凋亡过程中起着重要作用。
Objective: To investigate the effect of cyclooxygenase-2 inhibitor on angiogenesis in oral squamous cell carcinoma. Methods: Human tongue squamous cell carcinoma cell line Tca8113 was selected as the experimental sample. The effect of COX-2 inhibitor NS-398 on the mRNA and protein expression of VEGF and Survivin in Tca8113 cell line was detected by reverse transcription-polymerase chain reaction (PT-PCR) and Western blotting. SPSS11.5 software package for statistical analysis of the data. Results: NS-398 down-regulated the expression of VEGF and Survivin in Tca8113 cells and showed a time-dependent manner. CONCLUSION: NS-398, a COX-2 inhibitor, can reduce the mRNA and protein levels of Tca8113 VEGF and Survivin in oral squamous cell carcinoma and play an important role in angiogenesis and apoptosis in oral squamous cell carcinoma.
