目的筛选利福平依赖结核杆菌,并对其分子生物学特征进行初步鉴定。方法用含利福平(200μg/mL)的酸性罗氏培养基分离培养疑似结核病人的痰液,对筛选出菌株的rpoB基因进行测序分析。结果筛选出5株利福平依赖菌株,测序结果表明所有菌株在rpoB的核心突变区内均有突变发生。结论本研究筛选的利福平依赖菌株在ropB核心突变区内均有突变发生,但仍需进一步研究利福平依赖菌株的其他分子生物学特征。
Objective To screen for rifampin-dependent Mycobacterium tuberculosis and to identify its molecular biological characteristics. Methods The sputum of suspected tuberculosis patients was isolated and cultured with acidic Roche medium containing rifampicin (200μg / mL), and the rpoB gene of the selected strains was sequenced and analyzed. Results Five strains of rifampicin-dependent strains were screened. The sequencing results showed that all strains had mutations in the core mutation region of rpoB. Conclusion All of the rifampin-dependent strains screened in this study have mutations in the ropB core mutation region, but other molecular biological characteristics of rifampicin-dependent strains are still to be further studied.
