目的探讨干扰素(IFNs)对骨肉瘤细胞表达基质金属蛋白酶-2(MMP-2)和组织蛋白酶-D(Cath-D)的调控作用,并为临床上应用IFNs防治肿瘤转移提供一些实验基础。方法采用IFN-α及IFN-γ对体外培养的OS-732骨肉瘤细胞MMP-2和Cath-D表达进行诱导调节,并利用细胞酶联免疫吸附试验(C-ELISA)方法对其表达水平的变化进行半定量检测。结果IFN。及IFN-γ对MMP-2和Cath-D的表达均有显著的诱导降低作用(P<0.01),且诱导规律较为一致,即随IFNs浓度的增加其表达量随之逐渐下降。结论IFN-α及IFN-γ在一定程度上能抑制骨肉瘤的浸润转移潜能,在治疗骨肉瘤中有一定的积极作用。
Objective To investigate the regulatory effects of interferons (IFNs) on the expression of matrix metalloproteinase-2 (MMP-2) and cathepsin-D (Cath-D) in osteosarcoma cells, and to provide some experimental basis for the clinical application of IFNs in the prevention and treatment of tumor metastasis. Methods The expression of MMP-2 and Cath-D in OS-732 osteosarcoma cells cultured in vitro was induced by IFN-α and IFN-γ, and the expression level of MMP-2 and Cath-D was detected by using cell enzyme-linked immunosorbent assay (C-ELISA). Changes are semi-quantitative. Results IFN. Both IFN-γ and IFN-γ had significant effects on the induction of MMP-2 and Cath-D expression (P<0.01), and the induction law was consistent. That is, the expression level of MMP-2 and Cath-D gradually decreased with the increase of IFNs concentration. Conclusion IFN-α and IFN-γ can inhibit the invasive and metastatic potential of osteosarcoma to a certain extent, and have certain positive effects in the treatment of osteosarcoma.
