本研究采用体内实验与体外实验方法观察了DMF对大鼠肾脏钙稳态的影响。体内实验结果表明,DMF可明显抑制肾脏Ca ̄(2+)+ATPase活力,并可使肾脏磷酸化酶a活力升高。组织病理及超微结构观察表明DMF主要损伤肾小管上皮细胞。体外实验结果在明,DMF能明显抑制肾脏线粒体、微粒体对 ̄(45)Ca的主动摄取能力,并存在剂量-效应关系。以上结果提示在DMF肾脏损伤机理中钙稳态失调起重要作用。
In this study, in vivo experiments and in vitro experimental method to observe the effects of DMF on the calcium homeostasis in rats. In vivo experimental results show that DMF can significantly inhibit the renal Ca ~ (2 +) + ATPase activity, and renal phosphorylase activity increased. Histopathological and ultrastructural observation showed that DMF mainly damaged renal tubular epithelial cells. The results of in vitro experiments show that DMF can significantly inhibit the uptake capacity of mitochondria and microsomes in kidney, and there is dose-effect relationship. The above results suggest that calcium homeostasis plays an important role in the mechanism of DMF kidney injury.
