目的探讨非小细胞肺癌(NSCLC)SIRT6基因的表达并分析其与临床因素的相关性。方法收集2011年5月至2014年12月手术治疗、术后病理诊断为Ⅰ~Ⅲ期NSCLC患者38例为研究对象(研究组),其中腺癌20例,鳞癌18例,其中16例患者癌旁正常肺组织作为对照组。应用实时定量PCR、Western blot、免疫荧光检测腺癌、鳞癌癌组织及正常肺组织中SIRT6 mRNA蛋白的表达,对SIRT6蛋白的表达与NSCLC临床因素做Spearman相关分析。结果研究组腺癌及鳞癌肺癌组织SIRT6 mRNA及蛋白相对表达量均较对照组明显减少,差异有统计学意义(P均<0.01),而腺癌及鳞癌组织SIRT6 mRNA及蛋白相对表达量差异无统计学意义(P>0.05);正常肺组织可见到大量的SIRT6阳性细胞位于细胞核中,腺癌及鳞癌肺癌组织中几乎未见SIRT6阳性细胞,SITRT6阳性细胞百分比与正常肺组织比较差异有统计学意义(P均<0.01),而腺癌及鳞癌组织比较无统计学差异(P>0.05);SIRT6蛋白表达与非小细胞肺癌组织分期及分化程度无相关性(P均>0.05)。结论 NSCLC癌组织中几乎不表达SIRT6,SIRT6蛋白表达与非小细胞肺癌组织分期及分化程度无关,SIRT6基因可能在NSCLC发生的起始阶段发挥重要作用,但与疾病的进展无关。
Objective To investigate the expression of SIRT6 gene in non-small cell lung cancer (NSCLC) and analyze its relationship with clinical factors. Methods Thirty-eight patients with stage Ⅰ-Ⅲ NSCLC who were surgically treated from May 2011 to December 2014 were enrolled in the study. Among them, 20 were adenocarcinoma and 18 were squamous cell carcinoma. Among them, 16 patients Adjacent normal lung tissue as a control group. The expression of SIRT6 mRNA in adenocarcinoma, squamous cell carcinoma and normal lung tissues was detected by real-time quantitative PCR, Western blot and immunofluorescence. Spearman correlation analysis was performed between the expression of SIRT6 protein and clinical factors of NSCLC. Results The relative expression of SIRT6 mRNA and protein in lung adenocarcinoma and squamous cell carcinoma of the study group was significantly lower than that of the control group (P <0.01), while the relative expression of SIRT6 mRNA and protein in adenocarcinoma and squamous cell carcinoma There was no significant difference between the two groups (P> 0.05). SIRT6 positive cells were found in the nucleus in normal lung tissues. There was almost no SIRT6 positive cells in adenocarcinoma and squamous cell lung cancer tissues. The percentage of SITRT6 positive cells was significantly lower than that in normal lung tissues (P <0.01), but there was no significant difference between adenocarcinoma and squamous cell carcinoma (P> 0.05). There was no correlation between the expression of SIRT6 protein and the staging and differentiation of non-small cell lung cancer ). Conclusion SIRT6 and SIRT6 protein expressions in NSCLC tissues are not correlated with the staging and differentiation of non-small cell lung cancer. SIRT6 gene may play an important role in the initial stage of NSCLC, but not with the progression of the disease.
