目的:研究复元胶囊对大鼠实验性骨关节炎(OA)软骨白细胞介素-6(IL-6)和碱性成纤维细胞生长因子(bFGF)表达的影响。方法:Hulth法复制大鼠OA模型,随机分为对照、模型、四环素和复元胶囊高、中、低剂量组。灌胃给药,每天1次,连续10周。10周后取软骨标本,行大体观察、Mankin’s评分及免疫组化测定软骨中IL-6、bFGF的表达水平,并将IL-6、bFGF的表达水平与Mankin’s评分指数进行Pearson相关分析。结果:复元胶囊各组关节软骨Mankin’s评分、IL-6表达水平均低于模型组(P<0.01或P<0.05),bFGF表达水平均高于模型组(P<0.01);复元胶囊各剂量组Mankin’s评分差异具有显著性(P<0.05),IL-6表达水平与关节病理改变的Mankin’s评分指数呈正相关(P<0.01),bFGF表达水平与关节病理改变的Mankin’s评分无明显相关。结论:复元胶囊能下调大鼠OA软骨IL-6的表达,上调bFGF的表达,其作用呈剂量依赖性。高剂量复元胶囊抗大鼠OA作用与四环素相当。
Objective: To study the effect of Fuyuan Capsule on expression of interleukin-6 (IL-6) and basic fibroblast growth factor (bFGF) in cartilage of rats with experimental osteoarthritis (OA). Methods: The rat model of OA was duplicated by Hulth method and randomly divided into control, model, tetracycline and Fuyuan capsule high, medium and low dose groups. Gavage, 1 day, for 10 weeks. After 10 weeks, cartilage specimens were taken for gross observation. Mankin’s score and immunohistochemistry were used to determine the expression of IL-6 and bFGF in cartilage. Pearson correlation analysis was used to analyze the expression of IL-6, bFGF and Mankin’s score. Results: Mankin’s score and IL-6 expression of articular cartilage in each group were lower than those in model group (P <0.01 or P <0.05), and bFGF expression levels were higher in model group (P <0.01) The Mankin’s score was significantly different (P <0.05). The level of IL-6 was positively correlated with Mankin’s score of joint pathological changes (P <0.01). The expression of bFGF had no significant correlation with Mankin’s score of pathological changes. CONCLUSION: Fuyuan Capsule can down-regulate the expression of IL-6 in OA cartilage and increase the expression of bFGF in a dose-dependent manner. The effect of high-dose Fuyuan capsule on rat OA is comparable to that of tetracycline.
