目的调查精神分裂症患儿神经营养素-3基因(NT-3)甘氨酸(Gly)/谷氨酸(Glu)多态性与脑室扩大及微小躯体异常(MPAs)之间的关系。方法采用聚合酶链反应-限制性片段长度多态性方法分析218例精神分裂症患儿和210例健康儿童NT-3基因Gly/Glu多态性;采用CT测量患儿的脑室参数(计算哈氏值、三脑室宽度、脑室指数、侧脑室体部指数、侧脑室宽度指数、前角指数表示),采用waldrop编制的躯体异常量表(WS)修改版评定MPAs发生情况;采用方差分析、协方差分析、t检验比较NT-3不同基因型(Gly/Gly、Gly/Glu、Glu/Glu)患儿之间脑室值的差异,采用非参数检验及t检验比较不同基因型患儿之间的WS评分差异,脑室值与WS评分的相关关系采用Spearman相关分析。结果 1.研究组NT-3基因Gly/Gly频率高于对照组,差异有统计学意义(75.2%vs64.3%,χ2=6.86,P<0.05),等位基因Gly频率高于对照组,差异有统计学意义(86.2%vs81.0%,χ2=4.37,P<0.05)。2.不同基因型患儿之间各脑室值比较差异均无统计学意义(Pa>0.05)。3.Glu/Glu基因型患儿头部WS评分显著高于Gly/Gly和Gly/Glu型患儿(Z=2.237,2.728,Pa<0.05)。4.WS总分与哈氏值及三脑室宽度均呈正相关(r=0.26、0.29,Pa<0.01),与前角指数呈负相关(r=-0.178,P<0.05)。结论精神分裂症患儿NT-3基因Gly/Glu多态性与微小躯体异常有关,与脑室扩大之间无显著关系。
Objective To investigate the association of NT-3 Gly / Glu polymorphism in patients with schizophrenia with ventricular enlargement and micro-body abnormalities (MPAs). Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to analyze the Gly / Glu polymorphism of NT-3 gene in 218 schizophrenic children and 210 healthy children. The ventricular parameters were calculated by CT (P <0.05), the value of RV, the width of the third ventricle, the index of the ventricle, the index of the lateral ventricle, the index of the lateral ventricle and the anterior horn index). The incidence of MPAs was assessed by Waldrop’s modified version of the somatic abnormality scale (WS) Variance analysis and t test were used to compare the differences of ventricular volume between children with different genotypes of NT-3 (Gly / Gly, Gly / Glu, Glu / Glu). The non-parametric and t- WS score differences, ventricular volume and WS score correlation using Spearman correlation analysis. The frequency of Gly / Gly in NT-3 gene was higher in the study group than in the control group (75.2% vs64.3%, χ2 = 6.86, P <0.05) The difference was statistically significant (86.2% vs81.0%, χ2 = 4.37, P <0.05). There was no significant difference in ventricular values between children with different genotypes (Pa> 0.05). The head WS score of Glu / Glu genotype was significantly higher than that of Gly / Gly and Gly / Glu children (Z = 2.237,2.728, Pa <0.05). The total score of WSS was positively correlated with Hash value and width of the third ventricle (r = 0.26,0.29, Pa <0.01), but negatively correlated with anterior horn index (r = -0.178, P <0.05). Conclusion Gly / Glu polymorphism of NT-3 gene in children with schizophrenia is related to minor somatic abnormalities, but not to ventricular enlargement.
