目的探讨CD146在骨肉瘤患者组织中的表达情况,并分析其与骨肉瘤临床特征的关系。方法采用免疫组化法检测50例骨肉瘤患者组织中CD146的表达情况,并检测用CD34标记的肿瘤微血管密度MVD,并利用RT-qPCR和western blot方法检测骨肉瘤组织标本中CD146的表达。结果 CD146在骨肉瘤组织中过高表达,其MVD值显著高于骨肉瘤周围非恶性组织,CD146的表达与骨肉瘤的Enneking分期及有无肺转移显著相关(<0.01),随着骨肉瘤组织临床分期及组织分化的进展,CD146的表达明显提高,且骨肉瘤组织中有肺结转移组的阳性表达率显著高于无肺转移组。结论 CD146在骨肉瘤的发生和发展过程中起着十分重要的作用,其分子水平的高低可能可以成为衡量骨肉瘤疾病活动程度中的一个重要的指标。“,”Objective To investigate the expression of CD146 in osteosarcoma tissues and the relationship with clinical and pathological characteristics. Methods The expression of CD146 and microvessel density (MVD) in 50 cases of osteosarcoma tissues was determined by Immunohistochemistry, and further detecting the expression of CD146 in osteosarcoma tissues by RT-qPCR and Western blot. Results CD146 was overexpressed in Human Osteosarcoma Tissues, and the MVD value was significantly higher than nonmalignant tissue sur ounding in osteosarcoma, and the expression level of CD146 was cor elated with Enncking stage and pulmonary metastasis( <0.01). With the progress of clinical stage and histological dif erentiation of osteosarcoma, expression of CD146 was increased, and expression of CD146 in osteosarcoma with pulmonary metastasis group was significantly higher than those without pulmonary metastasis. Conclusion CD146 is related to the occur ence and development of osteosarcoma closely, and should play an important role in the process.
