背景:中医对类风湿关节炎rheumatoidarthritis,RA的认识和治疗积累()比较丰富的经验,尤其是经过近几年来的研究,取得了令人瞩目的进展,具有治疗方法多,疗效确切,毒副作用少的特点,因而从中医领域进一步探寻治疗RA更为有效的药物和方法,有着重要的科学意义和社会意义。而中药能否在RA发病中通过多途径、多靶点、多环节作用方式,抑制或阻止软骨和骨的破坏并促进他们的新陈代谢及其损伤的修复的研究更是少有报道。目的:探讨中药通痹灵抗软骨破坏的作用机制。设计:以实验动物为研究对象的完全随机设计,对照实验研究。单位:一所大学金匮教研室。材料:实验于2002-08/2003-12在广州中医药大学第一附属医院综合实验室完成。Wistar清洁级大鼠,雌性,体质量(110±20)g,35只。由解放军第一军医大学动物实验室提供,动物合格证号:2002A041。将Wistar大鼠适应性喂养3d后,随机分为正常组、造模组、通痹灵总碱组。通痹灵总碱由广州中医药大学热带病研究所植物化学分析室提取。方法:以胶原诱导型关节炎大鼠为动物模型,分组灌胃治疗后,做大鼠趾关节病理切片,常规苏木精-伊红染色,观察软骨病变;培养软骨细胞,以rrIL-1β体外刺激软骨细胞导致软骨细胞基质降解作为药理模型,加用不同浓度的通痹灵总碱,并以地塞米松作为对照组,?
BACKGROUND: Traditional Chinese medicine has accumulated a lot of experience in the recognition and treatment of rheumatoid arthritis rheumatoididarthritis (RA), especially after years of research. It has made remarkable progress, with many treatments, definite curative effect, and toxic and side effects. Due to its small number of features, it is of great scientific and social significance to further explore the more effective drugs and methods for the treatment of RA from the field of Chinese medicine. Whether traditional Chinese medicine can play a role in the pathogenesis of RA through multi-pathways, multi-targets, and multi-linkages, studies on the inhibition or prevention of destruction of cartilage and bone and the promotion of their metabolism and repair of their damage are rarely reported. Objective: To explore the mechanism of anti-cartilage destruction of Tongfuling. Design: Completely randomized design of experimental animals and controlled experimental studies. Unit: A university’s Jinyu Office. MATERIALS: The experiment was performed at the Comprehensive Laboratory of the First Affiliated Hospital of Guangzhou University of Chinese Medicine from August 2002 to December 2003. Wistar clean grade rats, female, body weight (110±20) g, 35 animals. Provided by Animal Laboratory of the First Military Medical University of PLA, animal certificate number: 2002A041. The adaptive feeding of Wistar rats for 3 days was randomly divided into normal group, modeling group and Tongluoling total base group. Tongzhuling total base was extracted from phytochemical analysis room of Institute of Tropical Diseases, Guangzhou University of Chinese Medicine. METHODS: Collagen-induced arthritis rats were used as an animal model. After sub-gastric administration, rat toe joints were pathologically sectioned, and routine hematoxylin-eosin staining was used to observe cartilage lesions. Cultured chondrocytes were treated with rrIL-1β in vitro. Stimulation of cartilage cells leads to matrix degradation of chondrocytes as a pharmacological model, plus different concentrations of TBA and dexamethasone as a control group.
