目的研究人结缔组织生长因子的透皮吸收。方法应用氯胺T法制备125I-人结缔组织生长因子(125I-rCTGF),采用家兔进行不同剂量的125II-rCTGF的完整皮肤和破损(烫伤)皮肤吸收试验。结果125I-rCTGF的比活度为2.0×105计数/(分.μg)。皮肤吸收试验表明,不同剂量125I-rCTGF透皮给药后家兔均未能通过皮肤经毛细血管吸收入血。结论为重组人CTGF临床试验提供必要的实验数据。
Objective To study the transdermal absorption of human connective tissue growth factor. Methods 125I-rCTGF was prepared by chloramine T method, and intact skin and damaged (scald) skin absorption test of 125II-rCTGF were performed in rabbits. Results The specific activity of 125I-rCTGF was 2.0 × 105 counts / (min. Μg). Skin absorption test showed that different doses of 125I-rCTGF rabbit after transdermal administration failed to be absorbed into the skin through the capillaries. Conclusion The experimental data for the clinical trial of recombinant human CTGF were provided.
