以吉林地区三种金针菇以及河南产地的金针菇为材料,测定、分析了金针菇rDNA-ITS序列,并通过与GenBank上已登录的金针菇ITS序列比对,采用邻接法(Neighbor Joining,NJ)构建系统发育树。结果表明,所有供试材料ITS全长在712 bp~719 bp范围内,G+C含量在49.4%-49.9%之间,各样品间有35个变异位点,13个信息位点,遗传距离在0~0.014之间,其中吉林地区3种金针菇样品部分位点发生突变,均没有同步进化。9个金针菇样品的ITS序列被分成了两大组,其余样品之间又出现很多分支,表明ITS可作为金针菇亲缘关系鉴定的分子标记。
Three kinds of Flammulina velutipes in Jilin area and Flammulina velutipes in Henan province were used as materials to determine and analyze the rDNA-ITS sequence of Flammulina velutipes. The sequence of rDNA-ITS was compared with ITS sequences of Flammulina velutipes registered in GenBank, and Neighbor Joining (NJ) tree. The results showed that the ITS length of all the tested materials ranged from 712 bp to 719 bp, the G + C contents ranged from 49.4% to 49.9%, with 35 variation loci, 13 information sites and genetic distance In the range of 0 ~ 0.014, some samples of Flammulina velutipes were mutated in Jilin area, and no synchronous evolution occurred. ITS sequences of nine Flammulina velutipes samples were divided into two groups, and many other branches appeared between the other samples, indicating that ITS could be used as a molecular marker to identify Flammulina.
