The authors confirm by in situ hybridization that the myeloperoxidase gene (MPO gene, a differentiation-specific gene) in patients with acute promyelocytic leukemia (APL) is easily located on chromosome 15 from chromosome 17. The expression of MPO is closely related to the differentiation of myeloid cells and is considered to be a meaningful marker of myeloid cells. The synthesis of MPO in myeloid cells occurs in the late myeloblasts and promyelocytes. In the diagnosis of acute leukemia, when the MPO of immature cells is higher than 3%, it is a reliable marker for determining the leukemia granulocyte-like cell line. In patients with acute promyelocytic leukemia, MPO has a high level of expression, while acute leukemia also has promyelocytic dysplasia. The cytogenetic marker of the disease is t (15;
