目的:探讨多囊卵巢综合征(PCOS)雌鼠预处理后胰腺组织匀浆中超敏C-反应蛋白(hs-CRP)、降钙素原(PCT)、白细胞介素-10(IL-10)与脂联素(ADP)水平变化以及胰腺超微结构改变。方法:建立PCOS雌鼠模型,将50只雌鼠随机分为5组(A组、B组、C组、D组和E组),分别予二甲双胍、吡格列酮以及两者联合干预,处死并解剖雌鼠,取胰腺组织,先留取小部分胰腺组织制片,在透射电镜(TEM)下观察胰腺超微结构的变化;余下胰腺组织制备成匀浆液行固相夹心法ELISA,检测hs-CRP、PCT、IL-10与ADP水平变化。结果:B组和D组胰腺组织匀浆液中hs-CRP水平高于A组、C组和E组,差异有统计学意义(P<0.05)。B组PCT水平高于A组、C组和E组,差异有统计学意义(P<0.05);D组PCT水平低于B组,差异有统计学意义(P<0.05);D组PCT水平高于E组,差异有统计学意义(P<0.05)。B组IL-10水平低于其余4组,差异有统计学意义(P<0.05)。B组ADP水平低于A组和E组,差异有统计学意义(P<0.05);C组ADP水平低于E组,差异有统计学意义(P<0.05);D组ADP水平低于B组和C组,差异有统计学意义(P<0.05)。A组和E组胰腺外分泌腺细胞超微结构均表现正常,B组外分泌腺细胞呈凋亡改变,C组和D组较B组有改善。结论:PCOS雌鼠促炎因子与抑炎因子处于失衡状态,二甲双胍和吡格列酮联合干预可能是纠正这种状态的最佳用药方式。
Objective: To investigate the expression of hs-CRP, PCT, IL-10 in pancreatic tissue after pretreatment of PCOS female rats. And adiponectin (ADP) levels and pancreatic ultrastructure changes. Methods: Fifty female mice were randomly divided into 5 groups (group A, group B, group C, group D and group E). Metformin and pioglitazone were respectively administrated. The level of PCT in group B was higher than that in group A, C and E (P <0.05); PCT in group D was lower than that in group B (P <0.05); PCT in group D Higher than E group, the difference was statistically significant (P <0.05). The level of IL-10 in group B was lower than the other 4 groups (P <0.05). The level of ADP in group B was lower than that in group A and group E (P <0.05); the level of ADP in group C was lower than that in group E (P <0.05); the level of ADP in group D was lower than that in group B There was a significant difference between group C and group C (P <0.05). The ultrastructures of pancreatic exocrine gland cells in group A and group E were all normal. The exocrine glandular cells in group B showed apoptosis changes. The group C and group D improved compared with group B. CONCLUSION: The proinflammatory cytokines and anti-inflammatory cytokines in PCOS female rats are in an unbalanced state. The combination of metformin and pioglitazone may be the best way to correct this condition.
