研究抗肿瘤单克隆抗体相对应的肿瘤相关抗原,对鉴定抗肿瘤单克隆抗体的特异性程度、灵敏度、亲和性及通过分析相关抗原的表达,了解肿瘤发生、发展的相关关系和提供临床运用可行性依据等有着重要意义。我们在制备了特异性较好的抗人小细胞肺癌单抗4B_3、2F_7的基础上,从细胞、组织及蛋白水平研究了相对应的肿瘤相关抗原的表达行为及初步特性。研究发现这两株单抗所识别抗原分子量均在50kd,在小细胞肺癌细胞株或组织中明显表达,而在其它类型的肺癌中仅部份病例有表达现象,除肝癌外其它肿瘤和正常组织均无明显表达。
To study the relative tumor-associated antigens of anti-tumor monoclonal antibodies, to identify the specificity, sensitivity, and affinity of anti-tumor monoclonal antibodies, and to analyze the expression of related antigens, to understand the relationship between tumor occurrence and development, and to provide clinical applications. The feasibility basis is of great significance. Based on the specific anti-small cell lung cancer monoclonal antibodies 4B_3 and 2F_7, we studied the expression and preliminary characteristics of the corresponding tumor-associated antigens from the cell, tissue and protein levels. The study found that the molecular weights of the antigens recognized by these two monoclonal antibodies were all 50kd and were clearly expressed in small cell lung cancer cell lines or tissues. However, in some other types of lung cancer, only some cases had expression, and in addition to liver cancer, other tumors and normal tissues No obvious expression.
