目的 :探讨增殖相关基因cyclinD1、Rb在妊娠滋养细胞肿瘤中的表达变化。 方法 :采用免疫组化S P法 ,检测 2 0例葡萄胎、15例侵蚀性葡萄胎、15例绒毛膜癌组织中两种基因蛋白产物的表达。结果 :在恶性滋养细胞肿瘤组织中 ,cyclinD1的阳性表达率随临床期别的增高呈递增趋势 ,而Rb呈递减趋势 ,二者在Ⅲ期的阳性表达率与Ⅰ期及Ⅱ期相比差异均具有显著性(P <0 0 5 ) ;化疗可降低cyclinD1在恶性滋养细胞肿瘤组织中的阳性表达 ,而对Rb无影响。 结论 :cyclinD1的过表达 ,Rb的缺失在妊娠滋养细胞肿瘤的发展中可能有重要的生物学意义
Objective: To investigate the expression of cyclinD1 and Rb in gestational trophoblastic tumor. Methods: The expression of two gene proteins in 20 cases of hydatidiform mole, 15 cases of invasive hydatidiform mole and 15 cases of choriocarcinoma was detected by immunohistochemical SP method. Results: In malignant trophoblastic tumor, the positive expression rate of cyclinD1 increased with the clinical stage, while the Rb showed a decreasing trend. The positive expression rate of cyclinD1 in stage Ⅲ was significantly lower than that in stage Ⅰ and Ⅱ (P <0.05). Chemotherapy could reduce the positive expression of cyclinD1 in malignant trophoblastic tumor tissue, but had no effect on Rb. CONCLUSION: Overexpression of cyclinD1 and loss of Rb may have important biological implications in the development of gestational trophoblastic tumors
