目的:观察清热解毒方(GJD)对脓毒症模型大鼠的治疗作用并探讨其细菌学方面的机制。方法:采用盲肠结扎穿孔法制作大鼠脓毒症模型,实验分为假手术对照组、脓毒症模型组和GJD治疗组(生药24 g.kg-1),给予清热解毒方治疗后观察大鼠生存情况,同时使用细菌培养和实时定量PCR的方法对腹水和血标本进行细菌学分析。结果:大肠埃希菌、金黄色葡萄球菌等条件致病菌在脓毒症模型组动物腹水和血液中大量繁殖,在治疗组中上述细菌含量明显减少(P<0.01)。结论:条件致病菌是脓毒症模型大鼠的主要致病菌,GJD可以通过抑制致病菌的繁殖而对脓毒症起到治疗作用。
Objective: To observe the therapeutic effect of Qingre Jiedu Fang (GJD) on septic rat model and to explore its bacteriological mechanism. Methods: The model of septic rat was made by cecal ligation and puncture. The experiment was divided into sham-operation control group, sepsis model group and GJD treatment group (crude drug 24 g.kg-1), treated with Qingrejiedu Decoction The survival of rats was studied. Bacterial culture and real-time quantitative PCR were used to analyze the ascites and blood samples. Results: The pathogenic bacteria such as Escherichia coli and Staphylococcus aureus proliferated significantly in ascites and blood of sepsis model group, and the bacteria in the treatment group significantly decreased (P <0.01). Conclusion: The pathogenic bacteria are the main pathogenic bacteria in sepsis model rats. GJD can play a therapeutic role in sepsis by inhibiting the growth of pathogenic bacteria.
