目的研究ATP生物荧光法检测结果与试验菌菌落数的相关性,评价该方法的准确性和可靠性。方法分别用3种ATP生物荧光检测仪和菌落计数法检测金黄色葡萄球菌、大肠杆菌和白色念珠菌梯度稀释的菌悬液,构建数学模型,用残差图进行模型验证。分别用相关系数r和组内相关系数ICC评价准确性和可靠性。结果 3种检测仪检测试验菌在检测范围内均为直线模型较合适,3种试验菌的lg(菌落数)与3种检测仪lg(RLU)间的r均在0.9以上,BT-112D、hygiena System SURE Plus和Clean-Trace NGi的ICC分别为0.68、0.92和0.82。结论lg(RLU)与lg(菌落数)基本成直线正相关,ATP生物荧光法准确性较好,可靠性尚可,适合作为一种初筛、预警或现场教育方法应用于医疗机构、托幼机构、疫源地等物体表面、手卫生等清洁消毒效果监测。
Objective To study the correlation between the results of ATP biofluorescence and the number of colonies of test bacteria and evaluate the accuracy and reliability of the method. Methods Gradient dilutions of Staphylococcus aureus, Escherichia coli and Candida albicans were prepared by three kinds of ATP biofluorescence detector and colony counting method, respectively. The mathematical model was established and the model was validated by residual graph. The correlation coefficient r and the intraclass correlation coefficient ICC were respectively used to evaluate the accuracy and reliability. Results The linearity of the three test instruments in the detection range was better than that in the linear range. The r between the three kinds of test bacteria (lg (colony number) and three kinds of detector lg (RLU) was above 0.9) ICC for hygiena System SURE Plus and Clean-Trace NGi were 0.68, 0.92 and 0.82, respectively. Conclusions lg (RLU) and lg (number of colonies) are basically linearly and positively related to each other. The ATP biofluorescence method has good accuracy and reliability and is suitable as a primary screening, early warning or field education method for medical institutions, Agencies, foci and other objects surface, hand hygiene and other cleaning and disinfection effects monitoring.
