目的探讨细胞毒性T淋巴细胞相关抗原4(CTLA-4)基因外显子1的49位点A/G多态性与成人隐匿性自身免疫性糖尿病(latent autoimmune diabetes in adults,LADA)的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对23例LADA患者、34例2型糖尿病(T2DM)患者和33例健康对照进行CTLA-4A/G多态性检测。结果LADA患者CTLA-4基因外显子1G49等位基因频率显著高于正常对照组(P<0.01)和T2DM组(P<0.01),主要表现为基因型GG显著高于正常对照组(P=1.3×10-2)和T2DM组(P=3.4×10-2),基因型AG也明显高于正常对照组(P<0.01)和T2DM组(P=3.3×10-2),基因型AA显著降低正常对照组(P<0.01)和T2DM组(P<0.01)。结论CTLA-4基因A/G49多态性与LADA易感性相关。
Objective To investigate the relationship between polymorphism of 49 locus A / G of exon 1 of cytotoxic T lymphocyte associated antigen 4 (CTLA-4) gene and latent autoimmune diabetes in adults (LADA). Methods CTLA-4A / G polymorphism was detected in 23 patients with LADA, 34 with type 2 diabetes mellitus (T2DM) and 33 healthy controls by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) . Results The frequencies of the allele 1G49 of exon 1 of CTLA-4 gene in LADA patients were significantly higher than those in normal controls (P <0.01) and T2DM patients (P <0.01). The GG genotype was significantly higher than that in normal controls (P = 1.3 × 10-2) and T2DM group (P = 3.4 × 10-2), genotype AG was significantly higher than that of the normal control group (P <0.01) and T2DM group (P = 3.3 × 10-2) Significantly reduced the normal control group (P <0.01) and T2DM group (P <0.01). Conclusion The A / G49 polymorphism of CTLA-4 gene is associated with the susceptibility to LADA.
