目的:研究调肝化浊解毒方及其拆方对非酒精性脂肪性肝炎(NASH)大鼠的脂质代谢的调控机制。方法:大鼠60只,除空白组10只外,均采用高脂饲料饲喂养,8周后停用高脂饲料,随机分为5组:模型组、全方组、化浊方组、解毒方组及易善复组;空白及模型组予蒸馏水灌胃,余治疗组分别给予相对应药物灌胃,治疗4周后检测各项指标。结果:全方组及其拆方组肝组织NAS评分、血清转氨酶及肿瘤坏死因子α(TNF-α)水平显著下降(P<0.01,P<0.05);血清高密度脂蛋白(HDL)和二磷酸腺苷(ADP)水平显著升高(P<0.01,P<0.05);肝组织甘油三酯(TG)含量及烯醛(HNE)、丙二醛(MDA)水平显著降低(P<0.01,P<0.05);肝组织谷胱甘肽过氧化物酶(GSH-PX)水平显著提高(P<0.01)。结论:调肝化浊解毒方可能主要通过ADP调节脂质代谢和炎性因子分泌从而达到治疗NASH的作用。
Objective: To study the regulatory mechanism of Tiaogan Huazhuo Jiedu Fang and its disassembled formulas on lipid metabolism in nonalcoholic steatohepatitis (NASH) rats. Methods: Sixty rats were fed with high-fat diet except for 10 rats in the blank group. After 8 weeks, the high-fat diet was stopped and randomly divided into 5 groups: model group, Danfang group, Huazhuo Fang group, detoxification Square group and easy to reconciliation group; blank and model group was given distilled water, the remaining treatment group were given the corresponding drug gavage, the detection of indicators after 4 weeks of treatment. Results: The liver tissue NAS score, serum aminotransferase and tumor necrosis factor α (TNF-α) levels in all the groups and their subgroups were significantly decreased (P <0.01, P <0.05) (P <0.01, P <0.05). The level of triglyceride (TG) and the level of HNE and MDA in liver tissue were significantly decreased (P <0.01, P < P <0.05). The level of glutathione peroxidase (GSH-PX) in liver tissue was significantly increased (P <0.01). Conclusion: Tiaogan Huazhuo Jiedu decoction may regulate the lipid metabolism and inflammatory factor secretion mainly through ADP to achieve the effect of treating NASH.
