目的:对山药多糖水提醇沉工艺进行研究,得出山药多糖的最佳提取工艺。方法:以山药总多糖为指标,采用分光光度法,单因素考察及正交设计试验法,对料液比、提取温度、提取时间、提取次数进行考察;并对醇沉浓度、醇沉时间和醇沉次数进行考察。结果:水提正交试验结果表明,料液比1:10,温度100℃,时间4h,提取3次为最佳工艺条件。醇沉正交试验表明,醇沉后溶液浓度为80%,时间为12h,醇沉1次。结论:该实验为山药多糖提取工艺的确定提供依据。
OBJECTIVE: To study the process of water extraction and alcohol precipitation of yam polysaccharide, and get the best extraction technology of yam polysaccharide. Methods: The total polysaccharide of yam was used as index to investigate the ratio of solid to liquid, extraction temperature, extraction time and extraction times by spectrophotometry, single factor test and orthogonal design test. The effects of alcohol concentration, alcohol precipitation time and The number of alcohol sinks were investigated. Results: The results of orthogonal test showed that the ratio of solid to liquid was 1:10, temperature was 100 ℃, time was 4h and extraction time was 3 times. Alcohol precipitation orthogonal test showed that after the alcohol solution concentration of 80%, the time was 12h, alcohol Shen 1 times. Conclusion: This experiment provides a basis for the determination of the extraction process of yam polysaccharide.
