目的 建立大鼠血清大黄酸的高效液相分析法 ,测定不同次数口服给予脑溢安浸膏血清中大黄酸的含量。方法 大鼠单次给 2g脑溢安浸膏和 4d 7次给药 (1g/次 ,2次 /d) ,于不同时间心内取血用HPLC测定大黄酸含量 ;HPLC条件 :C18柱 ,以 0 .1 %磷酸 -甲醇 (3 0∶70 )为流动相 ,紫外检测波长 43 5nm。结果 单次与多次给药均在 1h给药后达到最高血药浓度 ,但单次给药血药浓度迅速下降 ,而多次给药可维持较长时间的血浓平台。大黄酸在 0 .0 3 2~ 4.8mg·L-1范围内呈线性 ,r=0 .999,阴性无干扰 ,精密度RSD为 1 .83 % ,加样回收率为 96.9%。结论 检测方法简便、灵敏、准确、稳定 ;本品以每日 2次 ,连续给药为宜
Objective To establish a HPLC method for determination of serum rhein in rats and determine the content of rhein in the serum of Nao Yi An extract administered orally at different times. Methods Rats were given 2g Nao Yi An extract and 4d 7 doses (1g/time, 2 times/d) for a single time. Blood samples were taken from the heart at different time to determine rhein content by HPLC. HPLC conditions: C18 column. 0.1% phosphoric acid-methanol (30:70) was used as the mobile phase and UV detection wavelength was 43 5 nm. Results The single-dose and multi-dose administration reached the highest plasma concentration after one hour of administration, but the single-dose plasma concentration rapidly decreased, while multiple administrations could maintain a longer time blood concentration platform. Rhein was linear in the range of 0.032-4.8 mg·L-1, r=0.999, negative without interference, precision RSD was 1.83%, and the recovery rate was 96.9%. Conclusion The detection method is simple, sensitive, accurate, and stable. This product should be given twice a day and continuous administration is appropriate.
