目的:采用高效液相色谱法(HPLC)测定辣椒粉中的碱性橙。方法:称取样品于50 ml塑料离心管中,加入10 ml提取液,超声、离心后,取上清液过膜进样。采用XDB-C18色谱柱(5μm,4.6×250 mm),流动相为乙腈+乙酸铵(75+25),流速1.0 ml/min,二极管阵列检测器检测。结果:碱性橙在1.0μg/ml~50.0μg/ml范围内线性良好(r=0.9992),且峰形较好,回收率为93.4%,精密度RSD=0.61%,最低检出限为0.3μg/ml。结论:高效液相色谱法测定辣椒粉中的碱性橙,方法定性准确、灵敏度高、回收率和重现性好,操作快速,结果准确可靠。
Objective: To determine the basic orange in paprika by high performance liquid chromatography (HPLC). Methods: Weigh samples in 50 ml plastic centrifuge tube, add 10 ml extract, ultrasound, after centrifugation, the supernatant was injected through the membrane. XDB-C18 column (5μm, 4.6 × 250 mm) was used. The mobile phase consisted of acetonitrile + ammonium acetate (75 + 25), flow rate 1.0 ml / min and detected by diode array detector. Results: The calibration curve of basic orange was good (r = 0.9992) in the range of 1.0μg / ml ~ 50.0μg / ml with a good peak shape, the recovery was 93.4%, the precision RSD was 0.61% and the minimum detection limit was 0.3 μg / ml. Conclusion: Determination of alkaline orange in paprika by high performance liquid chromatography has the advantages of high accuracy, high sensitivity, good recovery and reproducibility, fast operation and accurate and reliable results.
