目的:研究重组人肿瘤坏死因子α(TNF-α)对放烧复合伤小鼠骨髓有核细胞(BMNC)IL-3R的调节作用。方法:以放烧复合伤小鼠为模型,观察一次性静脉注入不同剂量的TNF-α对BMNC增殖的影响及10万UTNF-α对IL-3Rα表达及功能、BMNC数量的调节作用。结果:①静注不同剂量的TNF-α对放烧复合伤小鼠BMNC增殖的影响不一;②10万U的TNF-α1、3d上调放烧复合伤小鼠IL-3Rα的平均含量(对阳性率及功能无明显影响),而致BMNC数量明显降低;7d时恢复至对照组水平;③10万UTNF-α1、3d致放烧复合伤小鼠BMNC计数明显降低。结论:10万UTNF-α在体内虽能上调放烧复合伤小鼠IL-3Rα的平均含量而剌激造血,但可能因其同时激活造血调控网络整体效应却抑制骨髓造血。
Objective: To investigate the regulatory effect of recombinant human tumor necrosis factor-α (TNF-α) on bone marrow nucleated cells (BMNC) IL-3R induced by radiation and burn in mice. Methods: The effects of single intravenous infusion of different doses of TNF-α on the proliferation of BMNC and the regulation of the expression of IL-3Rα by 10 000 UTNF-α and the number of BMNC were observed in burn-burn combined injury model mice. Results: ①The effects of different doses of TNF-α on the proliferation of BMNC in burn-burn injury mice were different; ② The average content of IL-3Rα in burn-burn injury mice was increased by 100000 U TNF-α1,3d Rate and function had no significant effect), while the number of BMNC was significantly decreased; on the 7th, the level of BMNC was restored to that of the control group; CONCLUSIONS: Although 1 000 000 UTNF-α can up-regulate the average level of IL-3Rα in mice after burn-combined injury, it may stimulate hematopoiesis, but may inhibit bone marrow hematopoiesis by activating the overall effect of hematopoietic regulation network.
