目的:建立HPLC-荧光检测法同时测定大鼠血浆及脑组织中5种单胺类神经递质的方法。方法:采用Kromasil C18反相柱,流动相为甲醇∶水(92∶8),其中水相中每升含NaAc 0.1mol、EDTA-2Na 0.1mol,HAc1.44mL,流速1mL.min-1,发射波长330nm,激发波长290nm。结果:线性范围:5-羟色胺(5-HT)0.30~15.20ng,多巴胺(DA)1.09~54.45ng,去甲肾上腺素(NE)1.77~88.50ng,5-羟色胺酸(5-HTP)0.26~12.83ng,5-羟色胺-3-吲哚乙酸(5-HIAA)0.25~12.70ng,线性关系与精密度良好,血浆及脑组织中5种单胺类神经递质的回收率在88.7%~105.7%之间。结论:该法操作简单、快速、准确,可用于检测大鼠血浆及脑组织中5种单胺类递质的含量。
OBJECTIVE: To establish a method for simultaneous determination of 5 monoamine neurotransmitters in rat plasma and brain by HPLC-fluorescence detection. Methods: Kromasil C18 reversed-phase column was used. The mobile phase consisted of methanol: water (92: 8), containing 0.1 mol of NaAc, 0.1 mol of EDTA-2Na and 1.44 mL of HAc in the aqueous phase Wavelength 330nm, excitation wavelength 290nm. Results: The linear range was from 0.30 to 15.20 ng of 5-HT, 1.09 to 54.45 ng of DA, 1.77 to 88.50 ng of norepinephrine, 0.26 to 5-HTP, The linearity and precision of 5-hydroxytryptamine-3-indoleacetic acid (5-HIAA) from 0.25 to 12.70ng were good, and the recoveries of five monoamine neurotransmitters in plasma and brain tissue ranged from 88.7% to 105.7 %between. Conclusion: The method is simple, rapid and accurate and can be used to detect the content of five monoamine neurotransmitters in rat plasma and brain tissue.
