目的:研究不同白毛夏枯草提取物对肝癌细胞增殖的抑制作用及对其可能的作用机制进行初步的研究。方法:用白毛夏枯草水提取物(STW)、醇提物(CTW)、乙酸乙酯萃取部位(ZTW)分别作用于HepG2肝癌细胞,采用四甲基偶氮唑盐(MTT)法检测这3种提取物对HepG2肿瘤细胞增殖的抑制作用,观察不同浓度的白毛夏枯草水提物对肝癌细胞株HepG2的增殖影响。采用荧光定量PCR技术检测STW、CTW、ZTW处理后的肝癌细胞株中BAX和Bcl-2 mRNA的表达情况。结果:STW、CTW、ZTW均能显著抑制多种肿瘤细胞的增殖,与阴性对照组相比,其可在mRNA水平上抑制Bcl-2、同时促进BAX mRNA的表达,具有剂量依赖性。结论:白毛夏枯草提取物对肿瘤细胞具有明显的抑制作用,其抑瘤机制可能与抑制肿瘤细胞中Bcl-2、同时促进BAX mRNA的表达相关。
OBJECTIVE: To study the inhibitory effect of different Prunella vulgaris extracts on the proliferation of hepatocellular carcinoma cells and its possible mechanism. Methods: HepG2 hepatocarcinoma cells were treated with aqueous extract of ST, CTW and ZTW, respectively, and detected by MTT assay Three kinds of extracts on HepG2 tumor cell proliferation inhibition observed different concentrations of Prunella vulgaris aqueous extract on the proliferation of HepG2 cells. Fluorescent quantitative PCR was used to detect the expression of BAX and Bcl-2 mRNA in STW, CTW and ZTW treated hepatoma cell lines. Results: STW, CTW and ZTW could significantly inhibit the proliferation of various tumor cells. Compared with the negative control group, STW, CTW and ZTW could inhibit Bcl-2 at mRNA level and promote BAX mRNA expression in a dose-dependent manner. CONCLUSION: Prunella vulgaris extract has a significant inhibitory effect on tumor cells. The anti-tumor mechanism may be related to the inhibition of Bcl-2 and the expression of BAX mRNA in tumor cells.
