目的研究高糖对人内皮细胞内皮抑素(ES)mRNA及蛋白表达水平的影响及其意义。方法培养人脐静脉内皮细胞(HUVECs),分为正常糖(5.6mmol/L)对照组,高糖(11.2、22.mmol/L)组。培养24、48、72、96h后,收集各组不同作用时间点细胞,用RT-PCR法检测ES mRNA表达水平,Western blot法检测ES蛋白的表达。结果高糖对HUVECs中ES mRNA和蛋白表达水平的影响是双相的:短时间内起促进作用,具有时间依赖性;随作用时间延长转为抑制作用,22.4mmol/L糖培养HUVECs 96h时组ES mRNA和蛋白表达水平较对照组明显降低(P<0.05)。结论在糖尿病(DM)慢性病程中,ES表达的降低可能与DM血管病变的发生有关。
Objective To investigate the effect and significance of high glucose on endostatin (ES) mRNA and protein expression in human endothelial cells. Methods Human umbilical vein endothelial cells (HUVECs) were cultured and divided into normal glucose (5.6 mmol / L) control group and high glucose (11.2, 22 mmol / L) groups. After cultured for 24, 48, 72 and 96 hours, the cells in different time points were collected. The expression of ES mRNA was detected by RT-PCR and the expression of ES protein by Western blot. Results The effect of high glucose on the expression of ES mRNA and protein in HUVECs was biphasic: the effect was promoted in a short period of time and in a time-dependent manner. The effect of high glucose on the expression of ES mRNA and protein in HUVECs was inhibited by 22.4 mmol / L glucose for 96 h ES mRNA and protein expression levels were significantly lower than the control group (P <0.05). Conclusion The decreased expression of ES may play an important role in the pathogenesis of DM in the chronic course of diabetes mellitus (DM).
