目的 :检测凋亡相关蛋白BAG 1与BAD在肝外胆管癌中的表达 ,探讨BAG 1与BAD在肝外胆管癌细胞凋亡调控中的作用以及与胆管癌分化的关系 .方法 :采用免疫组织化学方法 (DAKOEnVisionTMSystems) ,结合计算机图像分析系统与凋亡细胞计数 ,在组织原位定量检测 4 2例肝外胆管癌的凋亡状况及BAG 1与BAD蛋白的表达水平 .结果 :在 2 4例高分化、13例中分化和 5例低分化的肝外胆管癌中 ,免疫组化定量检测显示BAG 1阳性染色的平均吸光度值分别为 0 .0 6 8+ - 0 .0 37,0 .112 + - 0 .0 5 7和 0 .15 2 + - 0 .0 6 2 ;BAD阳性染色的平均吸光度值分别为 0 .0 79+ - 0 .0 5 3,0 .138+ - 0 .0 5 8和 0 .16 5 + - 0 .0 6 1,HE染色计数典型凋亡细胞的指数分别为 1.1+ - 0 .5 ,1.7+ - 0 .3和 3.5 + - 1.3.BAG 1、BAD的表达水平及凋亡指数在高、中、低分化的肝外胆管癌中逐级增加 (P <0 .0 5 ) ,凋亡指数与BAD的表达呈正相关 (P <0 .0 1) ,表明凋亡相关的BAG 1和BAD蛋白的表达与肝外胆管癌的分化程度有关 ,且二者可能参与了其凋亡的调控 .结论 :在肝外胆管癌中 ,凋亡相关蛋白BAG 1和BAD的表达及凋亡指数与肿瘤的分化有关 .BAG 1和BAD均有可能参与其凋亡调控 .
OBJECTIVE: To detect the expression of BAG-1 and BAD in extrahepatic cholangiocarcinoma and to explore the role of BAG-1 and BAD in the regulation of apoptosis in extrahepatic cholangiocarcinoma and their relationship with the differentiation of cholangiocarcinoma.Methods: (DAKOEnVisionTMSystems), combined with computerized image analysis system and apoptotic cell count, in situ quantitative detection of apoptosis in 42 cases of extrahepatic cholangiocarcinoma tissue and BAG 1 and BAD protein expression levels.Results: In 24 cases In well differentiated, moderately differentiated 13 cases and poorly differentiated 5 cases of extrahepatic cholangiocarcinoma, immunohistochemistry quantitative detection showed that the average absorbance values of BAG 1 -positive staining were 0.06 8+ -0.07,01.112 + - 0 .0 5 7 and 0 .15 2 + - 0 .0 6 2; The average absorbance values of BAD positive staining were 0 .0 79 + - 0. 0 5 3,0. 138 + - 0. 05 8 and 0 .16 5 + - 0 .0 6 1, the index of typical apoptotic cells counted by HE staining were 1.1 + -0.5, 1.7 + -0.3 and 3.5 + -1.3, respectively. The expression of BAG1 and BAD The level of apoptosis and apoptosis index increased progressively in high, moderate and poorly differentiated extrahepatic cholangiocarcinoma (P <0.05), and the apoptosis index was positively correlated with the expression of BAD (P <0.01) The expression of BAG 1 and BAD proteins correlated with the degree of differentiation of extrahepatic cholangiocarcinoma and both of them may be involved in the regulation of apoptosis.Conclusion: The expressions of BAG 1 and BAD in extrahepatic cholangiocarcinoma And apoptosis index are related to tumor differentiation.BAG 1 and BAD are likely involved in the regulation of apoptosis.
