目的:研究芪玄抑甲宁对甲亢大鼠Th17细胞相关因子的调控影响。方法:将雌性SD大鼠随机分成空白组(生理盐水组)和模型组,以尾静脉注射小肠结肠炎耶尔森氏菌造模,模型组大鼠随机分为模型组、芪玄抑甲宁14.4 g/kg、7.2 g/kg、3.6 g/kg剂量组和甲巯咪唑组(3mg/kg)。用酶联免疫法检测大鼠血清中IL-17、IL-6、TGF-β水平,免疫组化法和Real-time PCR法检测甲状腺组织中IL-17、IL-17R和IL-17mRNA、IL-17RmRNA的蛋白表达。结果:芪玄抑甲宁(14.4 g/kg)可以明显降低甲亢大鼠血清中IL-17、IL-6和TGF-β的水平,回调甲亢大鼠甲状腺组织中IL-17、IL-17R和IL-17mRNA、IL-17RmRNA的蛋白表达水平。结论:芪玄抑甲宁对Th17细胞的相关因子具有调控影响,这可能是芪玄抑甲宁治疗甲亢的重要作用机制之一。
Objective: To study the effect of Qi Xuan Yin Naoning on Th17 cell related factors in hyperthyroidism rats. Methods: Female Sprague-Dawley rats were randomly divided into blank group (saline group) and model group, and were injected with Yersinia enterocolitica in tail vein to make model. Rats in model group were randomly divided into model group, 14.4 g / kg, 7.2 g / kg, 3.6 g / kg dose group and the methimazole group (3 mg / kg). Serum levels of IL-17, IL-6 and TGF-β were measured by enzyme-linked immunosorbent assay. IL-17, IL-17R and IL-17 mRNA in thyroid tissue were detected by immunohistochemistry and Real- -17R mRNA protein expression. Results: Astragaloside (14.4 g / kg) significantly decreased the levels of IL-17, IL-6 and TGF-β in the serum of hyperthyroid rats, and reversed the levels of IL-17, IL-17R IL-17 mRNA, IL-17R mRNA protein expression levels. CONCLUSION: Quxuanyuyinning has a regulatory effect on the Th17 cell-related factors, which may be one of the important mechanisms of Quxuangyuening’s treatment of hyperthyroidism.
