目的 :探讨大黄虫丸对大鼠肝星状细胞表达转化生长因子β1(TGF β1)的影响。方法 :用CCl4复合因素致大鼠肝纤维化模型 ,同时分别给予 3种剂量大黄虫丸治疗 ,免疫组化方法观察TGF β1、α 平滑肌肌动蛋白 (α SMA)的合成表达及纤维化程度分级。结果 :经给予大黄虫丸干预治疗常规剂量组大鼠TGF β1、α SMA表达与模型组比较差异无显著性 (P >0 0 5 ) ,但双倍剂量组和三倍剂量组TGF β1、α SMA在汇管区及不连续纤维间隔的表达明显减弱 (P <0 0 5 ,P <0 0 1)。结论 :大黄虫丸较大剂量能抑制肝星状细胞增殖和分泌TGF β1,减少胶原生成 ,从而减弱肝星状细胞的自分泌放大效应 ,这可能是大黄虫丸抗肝纤维化作用的主要机制之一。
Objective: To investigate the effect of Dahuangqichong Pill on the expression of transforming growth factor-β1 (TGF-β1) in rat hepatic stellate cells. METHODS: Rat model of hepatic fibrosis was induced by CCl4 complex factor. Three doses of rhubarb acium pills were given. Immunohistochemistry was used to observe the expression and fibrosis of smooth muscle actin (α SMA). Grading. RESULTS: There was no significant difference in the expression of TGFβ1 and α SMA between the rats in the conventional dose group treated with Dahuangqichong Pill and the model group (P > 0.05), but the TGFβ1 in the double dose group and the triple dose group. The expression of α SMA in the portal area and discontinuous fiber septa was significantly decreased (P <0 05, P <0 01). Conclusion: Larger doses of rhubarb locust pills can inhibit the proliferation of hepatic stellate cells and secrete TGFβ1, reduce collagen production, and thus weaken the autocrine amplification effect of hepatic stellate cells. This may be due to the anti-fibrotic effect of rhubarb locust pills. One of the major mechanisms.
