本文通过建立Ex-vivo鼠胃腔模型,观察硝普钠(SodiumNitroprusside,Nitro)对内皮素(Endothelin,ET)所致的胃粘膜损伤后的损伤指数。光镜,扫描及透射电镜可见Nitro+ET组大鼠胃粘膜损伤程度比ET组轻;两组大鼠胃粘膜损伤指数有明显差异(P<0.001)。另外实验根据Fiske-Subbarow法,测得对照组、ET组、Nitro-ET组Na-K-ATP酶活力分别为:0.56±0.07、0.34±0.03、0.53±0.02,Nitro+ET组与ET组酶活力改变有显著差异(P<0.01),本实验表明:硝普钠具有减轻ET所致的胃粘膜损伤作用,并且可以抵消ET引起胃粘膜Na-K-ATP酶活力降低的影响。
In this study, we established gastric cavity model of Ex-vivo mouse to observe the injury index of gastric mucosal injury caused by sodium nitroprusside (Nitro) on endothelin (ET). Light microscope, scanning electron microscopy and transmission electron microscopy showed that the gastric mucosal lesion in Nitro + ET group was less than that in ET group. There was significant difference in gastric mucosal lesion index between the two groups (P <0.001). In addition, according to the Fiske-Subbarow method, the activity of Na-K-ATPase in control group, ET group and Nitro-ET group were measured as follows: 0.56 ± 0.07,0.34 ± 0.03,0.53 ± 0.02, Nitro + ET group and ET group were significantly different (P <0.01), this experiment shows that: sodium nitroprusside has the effect of reducing gastric mucosal damage caused by ET, and can counteract ET caused by gastric mucosal Na- K-ATP enzyme activity decreased.
