目的探讨血红素加氧酶-1对糖尿病大鼠神经病理性疼痛的影响及可能的机制。方法将造模成功的30只糖尿病大鼠随机分为3组,每组10只。甲组糖尿病鼠10只,乙组糖尿病鼠10只并采用锌原卟啉干预,丙组糖尿病鼠10只并采用钴原卟啉干预,另选取10只正常大鼠作为对照组。对所有大鼠在造模前和造模后进行测试大鼠的机械性缩足反应阈值(PWMT)。造模后43d,麻醉大鼠,取坐骨神经电镜标本;取L4~L6段脊髓,行HE染色,免疫组化染色及原位末端标记检测。结果在造模后7~42d内,甲乙丙3组的PWMT显著降低;与甲组相比,乙组的PWMT显著降低,丙组的PWMT显著升高,差异具有统计学意义(P<0.05)。结论血红素加氧酶-1对糖尿病大鼠神经病理性疼痛具有治疗作用,其可能的原因为血红素加氧酶-1能够减轻糖尿病外周神经病变及抑制脊髓背角神经元凋亡有关。
Objective To investigate the effect of heme oxygenase-1 on neuropathic pain in diabetic rats and its possible mechanism. Methods Thirty diabetic rats were randomly divided into three groups (n = 10 in each group). 10 diabetic rats in group A, 10 diabetic rats in group B were treated with zinc protoporphyrin, 10 diabetic rats were treated with cobalt protoporphyrin, and 10 normal rats were selected as the control group. The mechanical contracting threshold (PWMT) of the test rats was performed on all rats before and after modeling. At 43 days after model establishment, the rats were anesthetized and sciatic nerve electron microscopy was taken. The spinal cord of L4 ~ L6 segments was taken for HE staining, immunohistochemistry and in situ end labeling. Results Compared with group A, the PWMT of group B was significantly lower and the PWMT of group C was significantly higher than that of group A (P <0.05) . Conclusion Heme oxygenase-1 has a therapeutic effect on neuropathic pain in diabetic rats. The possible reason is that heme oxygenase-1 can reduce the peripheral neuropathy and inhibit the apoptosis of spinal dorsal horn neurons.
