Geranylgeranylacetone(GGA)为一种新的非环状的多类异戊二烯,是最近在日本发展的新抗溃疡药.Murakami等曾在实验性溃疡的研究中,证实它具有保护胃粘膜的作用,但其作用机制未明.本研究的目的是探讨GGA对鼠胃培养细胞有无刺激粘液合成和分泌的作用以及作用机制是否与前列腺素有关.所用动物系1~2周龄的Sprague-Dawley大鼠.取大鼠胃底全层组织作胃粘膜细胞培养,取培养3天后的胃细胞.用~3氢-葡萄胺渗入的方法进行粘液合成和分泌的测定.用放射免疫法测定培养基中前列腺素水平,主要为PGE_2和6-酮-PGF1α.所获结果均用统计学方法处理.结果:培养3天后细胞出现会合现象.90%以上的细胞被确定为能产生粘液的细胞.研究不同浓度GGA
