研究了建立草木樨优良悬浮系和单细胞培养再生植株的技术和适宜条件。由于叶和胚轴诱导的愈伤组织转入分化培养基中分化,取已分化出芽的愈伤组织和未分化的愈伤组织建立悬浮系,再由悬浮系分离出单细胞,单细胞经液体浅层培养得到了再生植株,试验结果表明,胚轴诱导的愈伤组织更适合于建立悬浮系和进行单细胞培养。在建立悬浮系过程中,每5天换一次新鲜培养基比较合适。由已分化出芽的愈伤组织建立的悬浮系分离的单细胞有较高的植株再生频率。
The technology and suitable conditions for establishing the excellent suspension system and the single-cell culture regenerated plant of Radix phellodendron were studied. Since the callus induced by leaves and hypocotyls was transferred to differentiation medium, the callus from the differentiated buds and the undifferentiated callus were used to establish a suspension system. Single cells were isolated from the suspension system. Single cells were separated by liquid The regenerated plants were obtained in shallow culture. The results showed that hypocotyl induced callus was more suitable for establishing suspension system and single cell culture. In the process of establishing a suspension system, a fresh medium every 5 days is more appropriate. Single cells isolated from suspension lines established by differentiated budding callus have a higher frequency of plant regeneration.
