目的分析微卫星不稳定性(MSI)在Ⅲ期结肠癌中导致的差异表达的肿瘤相关基因,以解释为何MSI与复发呈负相关。方法按特定筛选条件,搜寻NCBI公共基因表达数据库,使用R软件处理相关表达谱数据,包括统计学分析和差异基因分析。结果Ⅲ期结肠癌患者,在MSI未复发组与微卫星稳定性(MSS)复发组之间,通过差异基因表达分析,发现CAPN1、FLT3LG、SFN、ECT2、PTGS2、JUNB、ELF3、FOS、FOSB等基因在MSS复发组中高表达。PNAS4、ABCD3、CLDN8则在MSI未复发组中高表达。结论 PNAS4、ABCD3和CLDN8这3个基因的高表达,可能是MSI患者复发率低的原因。而MSS患者复发率高,可能是因为癌基因JUNB、FOS的高表达,促使细胞癌变转移。
Objective To analyze differentially expressed tumor-associated genes in microsatellite instability (MSI) in stage III colon cancer to explain why MSI was negatively correlated with recurrence. Methods According to the specific screening conditions, NCBI public gene expression database was searched, and related expression profile data were processed by R software, including statistical analysis and differential gene analysis. Results Among patients with stage Ⅲ colon cancer, CAPN1, FLT3LG, SFN, ECT2, PTGS2, JUNB, ELF3, FOS, FOSB, etc. were found by differential gene expression analysis between MSI non-recurrence group and microsatellite instability (MSS) The gene is highly expressed in the MSS recurrent group. PNAS4, ABCD3 and CLDN8 were overexpressed in MSI non-relapsing group. Conclusion The high expression of PNAS4, ABCD3 and CLDN8 may be the reason of low recurrence rate in MSI patients. The MSS patients with high recurrence rate may be due to oncogene JUNB, FOS high expression, prompting the metastasis of cancerous cells.
