目的通过对优选菌种长势较好7株子实体实施组织分离后菌丝生长情况的比较研究优选菌株最佳分离部位、菌丝生长适宜培养基,为北虫草菌种的复壮技术提供一定的科学依据。方法从分离菌株的成活率、污染率、分离菌株菌丝在不同培养基中的菌丝生长情况方面进行比较研究。结果北虫草组织分离可采用酒精浸泡15s,再用无菌水漂洗方法进行消毒;北虫草头可作为组织分离的最佳部位;综合培养基适宜于北虫草组织分离菌丝的生长。结论组织分离可作为获取北虫草优质菌种的一种有效的方法。
OBJECTIVE To study the mycelial growth of seven strains of fruiting bodies with better growth status and to choose suitable culture medium for mycelium growth to provide the best scientific basis for the rejuvenation of Cordyceps species in accordance with. Methods The mycelial growth of isolates was compared in terms of the survival rate, the contamination rate and the mycelium growth of the isolated strains. Results Cordyceps tissue could be soaked in alcohol for 15s, then disinfected by sterile water rinsing. Cordyceps militaris could be used as the best site for tissue separation. The synthetic medium was suitable for the growth of mycelium isolated from Cordyceps militaris. Conclusion Tissue isolation can be used as an effective method to obtain high quality strains of Cordyceps militaris.
