目的 观察异丙酚对大鼠心肌缺血/再灌注过程中骨形态构建蛋白-7基因(BMP-7)mRNA表达的影响。方法 Wistar大鼠30只,随机分成3组(n=10)。对照组(组Ⅰ)行假手术;缺血/再灌注组(组Ⅱ)可逆性阻断冠状动脉造成缺血10min后再灌注;异丙酚组(组Ⅲ)则在缺血过程中以1mg·kg-1·min-1静脉输注异丙酚。所有大鼠心肌组织标本行RT-PCR检查以确定BMP-7mRNA表达的改变。结果 组ⅠBMP-7 mRNA表达的丰度为1.14±0.08;组Ⅱ其表达有明显降低(丰度为0.90±0.05,P<0.05),组Ⅲ的BMP-7 mRNA表达(丰度为1.00±0.08)与组Ⅰ相比有明显降低(P<0.05),但与组Ⅱ比较明显改善(P<0.05)。结论 心肌缺血/再灌注损伤使BMP-7mRNA表达明显减少,提示BMP-7基因表达受到抑制。在缺血/再灌注过程中异丙酚通过改善BMP-7mRNA表达,在一定程度上减轻了心肌缺血/再灌注损伤。
Objective To investigate the effect of propofol on the expression of BMP-7 mRNA during myocardial ischemia / reperfusion in rats. Methods Thirty Wistar rats were randomly divided into three groups (n = 10). The rats in control group (group Ⅰ) underwent sham operation. The ischemic / reperfusion group (group Ⅱ) reversibly occluded the coronary artery and then reperfused 10 minutes after ischemia. Propofol group (group Ⅲ) · Kg -1 min -1 intravenous propofol. All rat myocardial tissue samples were examined by RT-PCR to determine the change in BMP-7 mRNA expression. Results The mRNA expression level of BMP-7 in group Ⅰ was 1.14 ± 0.08. The expression of BMP-7 in group Ⅱ was significantly lower than that in group Ⅱ (abundance was 0.90 ± 0.05, P <0.05) ) Was significantly lower than that of group I (P <0.05), but significantly improved compared with group II (P <0.05). Conclusion Myocardial ischemia / reperfusion injury significantly reduced the expression of BMP-7 mRNA, suggesting that BMP-7 gene expression was inhibited. Propofol relieved myocardial ischemia / reperfusion injury to a certain extent by improving BMP-7mRNA expression during ischemia / reperfusion.
